Rapamicina

IMPORTANT: Rapamycin is not a peptide; it is a macrolide (natural compound produced by the bacterium Streptomyces hygroscopicus, discovered in soil from Easter Island — Rapa Nui). Originally developed as an immunosuppressant for transplants, it is now the most promising substance in longevity research — the only drug consistently proven to extend lifespan in mammals.

Mechanism of Action

Rapamycin inhibits the mTOR protein (mechanistic Target Of Rapamycin), a central regulator of cell growth, metabolism, and autophagy. Partial mTOR inhibition activates autophagy (cellular recycling), mimics the effects of fasting and caloric restriction, and has anti-aging effects across multiple tissues.

Protocolo de Matt Kaeberlein (Dog Aging Project)

Inibidor de mTOR. Dose baixa intermitente melhora resposta imune em idosos. Doses altas são imunossupressoras (transplante). Uso anti-aging é experimental.

Benefits

Increased lifespan in mammals

Proven

The only compound consistently proven to increase lifespan in mice, rats, and other animal models. 9-26% lifespan increase in mice.

Immunosuppression (approved use)

Proven

Approved for organ transplant rejection prevention. The only officially approved use.

Immune function in the elderly

Under research

The PEARL study and others showed that low intermittent doses improve immune response to vaccines in the elderly without causing significant immunosuppression.

Human longevity

Unproven

Despite animal evidence, the effect on human longevity has not yet been demonstrated in clinical trials. Studies are underway.

Risks and Side Effects

Immunosuppression

High(Dose-dependent)

At continuous doses (transplants), it suppresses the immune system, increasing the risk of infections and cancer. Low intermittent doses for longevity carry lower but not negligible risk.

Metabolic disturbances

Moderate(Common)

May cause insulin resistance, dyslipidemia, and glucose intolerance, paradoxically increasing the risk of diabetes.

Mouth sores and mucositis

Low(30%)

Oral ulcers are a common side effect, even at low doses.

Off-label use

Moderate(N/A)

Use for longevity is off-label (outside the approved indication). Requires medical prescription and monitoring. Not sold over the counter.

Internet vs. Science

What people say online compared to the actual scientific evidence.

Longevity pill

Partially true

What they claim

Rapamycin is the pill that extends life — approved by longevity doctors.

Actual evidence

It is the most promising compound in animal longevity research, with solid evidence in mice. But human evidence is still limited and there are real risks. It is not a consequence-free 'youth pill'.
